Continued approval for this indication may be contingent upon verification and description of clinical benefit in confirmatory trial(s).<\/p>\n<h2 class=\"wp-block-heading\">DOSAGE AND ADMINISTRATION:<\/h2>\n<p class=\"wp-block-paragraph\">* Confirm the presence of an FGFR2 fusion or rearrangement prior to initiation of treatment with LuciNfig.<\/p>\n<p class=\"wp-block-paragraph\">* Recommended dosage: 125 mg orally once daily for 21 consecutive days followed by 7 days off therapy, in 28-day cycles.<\/p>\n<p class=\"wp-block-paragraph\">* Take on an empty stomach at least 1 hour before or 2 hours after food, at approximately the same time each day.<\/p>\n<p class=\"wp-block-paragraph\">* Swallow capsules whole with a glass of water. Do not crush, chew or dissolve.<\/p>\n<p class=\"wp-block-paragraph\">* Mild and Moderate Renal Impairment: The recommended dosage is 100 mg orally once daily for 21 consecutive days followed by 7 days off therapy, in 28-day cycles.<\/p>\n<p class=\"wp-block-paragraph\">* Mild Hepatic Impairment: The recommended dosage is 100 mg orally once daily for 21 consecutive days followed by 7 days off therapy, in 28-day cycles.<\/p>\n<p class=\"wp-block-paragraph\">* Moderate Hepatic Impairment: The recommended dosage is 75 mg orally once daily for 21 consecutive days followed by 7 days off therapy, in 28 day cycles<\/p>\n<h2 class=\"wp-block-heading\">WARNINGS AND PRECAUTIONS:<\/h2>\n<p class=\"wp-block-paragraph\">* Ocular Toxicity LuciNfig can cause retinal pigment epithelial detachment (RPED). Perform comprehensive ophthalmic examination including optical coherence tomography (OCT) prior to initiation of LuciNfig and at 1 month, at 3 months, and then every 3 months thereafter during treatment. Withhold as recommended.<\/p>\n<p class=\"wp-block-paragraph\">* Hyperphosphatemia and Soft Tissue Mineralization: Increases in phosphate levels can cause hyperphosphatemia leading to soft tissue mineralization, cutaneous calcinosis, non-uremic calciphylaxis, vascular calcification, and myocardial calcification. Withhold, dose reduce, or permanently discontinue as recommended.<\/p>\n<p class=\"wp-block-paragraph\">* Embryo-Fetal Toxicity: Can cause fetal harm. Advise patients of reproductive potential of the potential risk to the fetus and to use effective contraception.<\/p>\n<h2 class=\"wp-block-heading\">ADVERSE REACTIONS:<\/h2>\n<p class=\"wp-block-paragraph\">* Most common (\u226520%) adverse reactions were nail toxicity, stomatitis, dry eye, fatigue, alopecia, palmar-plantar erythrodysesthesia syndrome, arthralgia, dysgeusia, constipation, abdominal pain, dry mouth, eyelash changes, diarrhea, dry skin, decreased appetite, vision blurred and vomiting.<\/p>\n<p class=\"wp-block-paragraph\">* Most common laboratory abnormalities (\u226520%) were increased creatinine, increased phosphate, decreased phosphate, increased alkaline phosphatase, decreased hemoglobin, increased alanine aminotransferase, increased lipase, increased calcium, decreased lymphocytes, decreased sodium, increased triglycerides, increased aspartate aminotransferase, increased urate, decreased platelets, decreased leukocytes, decreased albumin, increased bilirubin and decreased potassium.<\/p>\n<h2 class=\"wp-block-heading\">DRUG INTERACTIONS:<\/h2>\n<p class=\"wp-block-paragraph\">* Strong or Moderate CYP3A Inhibitors: Avoid coadministration.<\/p>\n<p class=\"wp-block-paragraph\">* Strong or Moderate CYP3A Inducers: Avoid coadministration.<\/p>\n<p class=\"wp-block-paragraph\">* Gastric Acid Reducing Agents: Avoid coadministration. If coadministration cannot be avoided, stagger administration of LuciNfig from H2 antagonist or locally-acting antacid.<\/p>\n<h2 class=\"wp-block-heading\">USE IN SPECIFIC POPULATIONS:<\/h2>\n<p class=\"wp-block-paragraph\">Lactation: Advise not to breastfeed.<\/p>\n<h2 class=\"wp-block-heading\">Storage:<\/h2>\n<p class=\"wp-block-paragraph\">Store at 20\u2103 to 25\u2103 (68\u2109 to 77\u2109), excursions permitted between 15\u2103 and 30\u2103 (59\u2109 and 86\u2109) [see USP Controlled Room Temperature].
